Figure 1: BBL Enterotube II code sheet used for the identification of Gram (-), oxidase (-), Enterobacteriacae. BBL Enterotube II code sheet used for the identification of Gram (-), oxidase (-), Enterobacteriacae. Once the tube is inoculated and incubated for 24 hrs at 37°C results are read. If the test is read as a negative then no value is assigned. If the test is read as a positive then the test is assigned a value as seen below the test on the code sheet. Values then are added up in each block. The resulting code is then checked in the BBL Interpretation Guide for organism identification. (Tasha Sturm, Cabrillo College, Aptos, CA).

Figure 3: Organism inoculated in the BBL Enterotube II. The following organism we inoculated in the BBL Enterotube II and incubated for 24 hours at 37°C. A) Proteus vulgaris, B) Enterobacter aerogenes, C) Escherichia coli, D) Salmonella arizonae, E) Serratia marcescens, F) Proteus mirabilis (Tasha Sturm, Cabrillo College, Aptos, CA).

Note: for student safety SIM tubes were also done and the indole test was done by adding Kovac's reagent to the SIM tube vs adding into the Enterotube via a syringe Results: Glucose (+) media turned yellow, gas was not seen (indicated by separation of the media), Lysine (-) media remained yellow, Ornathine (-) media remained yellow, Hydrogen sulfide (+) indicated by blackening of the media in both the SIM and Enterotube, Indole (+) red ring at top of SIM tube, Adonitol (-) media remained red, Lactose (-) media remained red, Arabinose (-) media remained red, Sorbitol (-) media remained red, Voges-Proskauer test not done, Dulcitol (-) Phenylalanine (+) media turned black, Urea (+) media turned pink, Citrate (-) media remained green. (Tasha Sturm, Cabrillo College, Aptos, CA).
